EGF开发是早在60年代初Montalcini 和Cohen教授在純化小鼠頜下腺神經生長因數(NGF)時發現一組可促進新生小鼠提早開眼、長牙且對熱穩定的多肽類物質。最早發現它具有搞胃酸分泌作用,故又稱抑胃素。隨後將這一活性組份加入培養的皮膚表皮時發現,它可直接促進表皮生長,為此而定名為表皮生長因數(Epidermal growth factor,EGF)。1974年從人尿中提純出人的表皮生長因數(hEGF),其結構由53個氨基酸組成,分子量6201道爾頓,分子內有6個半胱氨酸組成的二硫鍵,形成3個分子內環型結構,組成生物活性所必須的受體結合區域。EGF無糖基部位,非常穩定,耐熱耐酸,廣泛存在於體液和多種腺體中,主要由頜下腺、十二指腸合成,在人體的絕大多數體液中均已發現,在乳汁、尿液、精液中的含量特異性地增高,但在血清中的濃度較低。眾多的實驗研究表明,EGF可刺激多種細胞的增殖,主要是表皮細胞、內皮細胞。用於角膜損傷、燒燙傷及手術等創面的修復和癒合取得了很好的療效,Montalcini 和Cohen教授因為發現表皮生長因數並分析其結構和作用機理,1986年諾貝爾生理學及醫學獲獎。
